I changed the belt and sliders on a 50cc Milano. I peg out at max speed but the take off is incredibly slow. Could this be due to over torquing the variator?

Varo weights:
Lighter weights = fast accl, lower top speed
Heavier weights = higher top speed, slower take off

Your weights are too heavy. Usually we reduce the weight of the rollers on 50cc's to get better acceleration.
No, if you variator is over-tightened, the only problem you will have is it's going to be difficult to remove the variator lock nut next time. The crankshaft has a shoulder machined onto it, preventing this sort of thing.
Try 6 g weights or 7g sliders
